Project context and summary:

Shank2- and Shank3-deficient mice are two mouse models of autism. We previously validated the use of a kit to extract both RNA and protein from the same sample with a quality suitable for transcriptomic and proteomic analysis, respectively. It was done on extracts of the striatum from WTs and Shank3-deficient mice. We now want to perform the same kind of analysis on multiple brain areas (Striatum, Cortex, Thalamus and cerebellum) in 4 males and 4 females Shank2 WT, 4 males and 4 females Shank2 KO, 4 males and 4 females Shank3 WT and 4 males and 4 females Shank3 KO. We want to investigate the differentially expressed genes in both models, if there is a sex effect, and see if we can correlate those results with results from a proteomic analysis that will be performed on the same samples in parallel (we use the Norgen RNA/Protein purification plus kit).
