We develop a technology to screen pooled library of regulated yeast strains with CRISPRa/i system to produce a protein of interest, relying on the enrichment of good clones throughout the culture. This enrichment is detected using deep sequencing to quantify the presence of each gRNA present at early and late time of the culture.
