Project context and summary:

G4 structures are non-canonical nucleic acid structures regulating gene expression and chromatin organization. These structures also regulate the replication of several viruses such as HIV-1. The aims of this project are to identify specific G4 ligands interacting with G4s present in HIV-1 LTR promoter and to study their effects on viral expression and chromatin organization surrounding the integrated viral genome. For this purpose, we will perform both transcriptomic and chromatin conformation capture studies in cell lines containing a silent and reactivable integrated HIV-1 genome, in the presence and absence of selected G4-ligands. These studies should reveal new mechanisms regulating HIV-1 expression that could be used as antiviral targets.
