BRCA1 and BRCA2 deficiencies are associated with several cancers and render cells hypersensitive to PARP inhibitors. How these cells survive and proliferate despite compromised DNA repair capacity remains largely unexplored. Here, we performed genome-wide screens in BRCA1/2 deficient cells and mapped their synthetic lethal interactions. This map provides a rich resource to study the mechanisms of cellular adaptation to BRCA1 and BRCA2 deficiencies.
